I agree that X128 is a great DOS emulator, unfortunately it seems to be a bit dodgy under Windows XP. As long as you can run DOS programs through Windows (I think Windows 2000 won't allow it, you see) X128 is the best bet. It will open most Speccy files and seems highly accurate. The latest version of X128 has ZX81 support, although its only at beta stage. But maybe it's possible to save files to tzx from there.
I'm not sure myself, as I have no ZX81 software to try that with.

X128 has mouse emulation, as well as offering almost everything else you ask for in a Spectrum emulator. I recommend using it and SGD (both available from this site) if you don't mind DOS programs.
